abstract
objective(s)para-nonylphenol (p-np) is used in many industries and our previous study showed that p-np causes a reduction in rats bone marrow mesenchymal stem cells (mscs) viability. the aim of this study was to investigate the effect of p-np on osteogenic differentiation of mscs.materials and methodsmscs were isolated and expanded to 3rd passage, then cultured in dmem supplemented with osteogenic media as well as 0.5 or 2.5 μm of p-np. after 5, 10, 15, and 21 days, the viability and the level of mineralization was determined using mtt assay and alizarin red, respectively. in addition, morphology and nuclear diameter of the cells were studied with the help of fluorescent dye. furthermore, calcium content and alkalinphosphatase activity were also estimated using commercial kits. data were statistically analyzed and the p<0.05 was taken as the level of significance.resultsthe viability and mineralization of the cells treated with 2.5 μm of p-np reduced significantly after day 10 in comparison with the control group and administration of 0.5 μm. moreover, chromatin condensation, reduction of nuclei diameter, and cytoplasm shrinkage was observed in the cell treated with 2.5 μm. the calcium concentration and alkalinphosphatase activity of the cells decreased significantly with 2.5 μm of p-np when compared with 0.5 μm and control group.conclusionadverse effect of p-np was observed on osteogenic differentiation of mscs at 2.5 μm due to disruption of mineralization. we strongly suggest more investigations on this chemical with respect to other stem cells, especially skin stem cells as p-np is used in the formulation of cosmetics.

full textSodium Arsenite Caused Mineralization Impairment in Rat Bone Marrow Mesenchymal Stem Cells Differentiating to Osteoblasts
ABSTRACT Background: Sodium arsenite (SA) recently has been recommended to be used in malignancy therapy. Our studies showed, SA in short and long period of treatment caused reduction of rats Bone Marrow Mesenchymal Stem Cells (MSCs) viability and induced caspase dependent apoptosis. The aim of this study was to investigate the effect of SA on osteogenic differentiation of MSCs. Methods: MSCs...
full textOsteogenic Differentiation of Rat Mesenchymal Stem Cells from Adipose Tissue in Comparison with Bone Marrow Mesenchymal Stem Cells: Melatonin As a Differentiation Factor
Background: Adipose-derived stem cells (ADSC) could be an appealing alternative to bone marrow stem cells (BMSC) for engineering cell-based osteoinductive grafts. Meanwhile, prior studies have demonstrated that melatonin can stimulate osteogenic differentiation. Therefore, we assayed and compared the melatonin effect on osteogenic differentiation of BMSC with that of ADSC. Methods: Mesenchymal...

Objective(s):Sea cucumber derived bioactive compound is considered efficient in treatment of bone disorders. This study was performed to evaluate the effect of this extract on differentiation of rat bone marrow mesenchymal stem cells (rBMMSc) into osteogenic lineage. Materials and Methods: Isolated rBMMSc were grown in DMEM supplemented with 10% FBS. The cells were exposed to different concen...
full textPara-nonylphenol Toxicity Induces Oxidative Stress and Arrests the Cell Cycle in Mesenchymal Stem Cells of Bone Marrow
Background: The mechanism of para-nonylphenol (PNP) reducing the proliferation and differentiation of bone marrow mesenchymal stem cells (MSCs) is not known. The present study was designed to investigate the mechanism. Methods: MSCs were extracted under sterile condition from Wistar rat and cultured in DMEM, containing 15 % FBS and penicillin/streptomycin until the 3rd passage, then cells we...
